The work performed during this training course is a preliminary step to autotroph respiration study (bon évidemment sans assurance aucune pour les termes techniques). Usually, in oceanic areas, respiration is determined since microb/germ population. Autotroph respiration is hard to monitor due to actual techniques. They only monitor whole population respiration. However, autotrph respiration has a role of prior importance in carbon cycle, then it is required to estimate it precisely.
We hypothetized that the latter respiration is made of a constant part depending on the biomass and a variable one depending on photosythesis.
Primary goal is to determine in vitro the Dunaliella tertiolecta respiration, single-celled chlorophycea, chosen for its cell culture caracteristics. This experimental data should have let us assess, among three differently formulated models for autotroph respiration, which model was the most representative, the one that would fits the best in situ and thus let us describe best this respiration.
Technical matters refrained us from carrying out these experiments, but we were at least able to make sure of signal stability from the cytometer (which let us built a curve), we were also able to detect initial source contamination and study respiration formalisation models
This training course let test the protocol, coordinate the different factors and identify problems so as to perform experiments in optimal conditions. We established as well that the 14C models were fine for our study, which requires short periods of study and vizualisation of iraadiancy effect on respiration.
Next step will be to carry out the inital project, which would logically follow the actual set up and determine estimation tools for autotrophe respiration.
